Influence of ROS on Ovarian Functions

چکیده
High level of ROS (Reactive Oxygen Species), due to an increased production of oxidant species and/or a decreased efficacy of antioxidant system, can lead to oxidative stress (OS) an emerging health risk factor involved in the aging and in many diseases, either in humans or in animals. ROS are a double-edged sword – they serve as key signal molecules in physiological processes, but also have a role in pathological processes involving the female reproductive tract. ROS affect multiple physiological processes in reproduction and fertility, from oocyte maturation to fertilization, embryo development and pregnancy. Several studies indicate that follicular atresia in mammalian species due to the accumulation of toxic metabolites often results from oxidative stress. It has been suggested that ROS under moderate concentrations play a role in signal transduction processes involved in growth and protection from apoptosis. Conversely, increase of ROS levels is primarily responsible for the alteration of macromolecules, such as lipids, proteins and nucleic acids, that lead to significant damage of cell structures and thereby cause oxidative stress. To prevent damage due to ROS, cells possess a number of non-enzymatic and enzymatic antioxidants. Non-enzymatic antioxidants include vitamin C, glutathione and vitamin E. Enzymatic antioxidants consist of superoxide dismutases (MnSOD and Cu/ZnSOD) that convert superoxide into hydrogen peroxide; glutathione peroxidase (GPX) and catalase (CAT) which neutralize hydrogen peroxide. Intracellular homeostasis is ensured by the complex interaction between pro-oxidants and

متن کاملEquilibrium between anti‐oxidants and reactive oxygen species: a requisite for oocyte development and maturation
Reactive oxygen species (ROS) are required for cellular functioning and are controlled by anti-oxidants. The ROS influence the follicles, oocytes, endometrium, and their environment. The luteinizing hormone surge initiates a massive recruitment of ROS that modulates major reproductive functions namely, oocyte maturation, ovarian steroidogenesis, corpus luteal function, and luteolysis. The anti-...
متن کاملP-224: The Effect of Alpha Lipoic Acid on ROS Production and TAC Levels of Cultured Isolated Pre-antral Follicles Drived from Mice Vitrified Ovaries
Background: Ovarian cryopreservation is an efficient way to preserve fertility of cancer patients before chemotherapy. This study was designed to evaluate the effect of apha lipoic acid (ALA) on reactive oxygen species (ROS) production and total antioxidant capacity (TAC) of cultured isolated pre-antral follicles derived from vitrified and non-vitrified ovarian tissue. Materials and Methods: Im...
متن کاملOxidative Stress and its Role in Female Infertility and Assisted Reproduction: Clinical Implications
Reactive oxygen species (ROS) are involved in physiological functions and act as mediators in various signaling processes. Elevated or sustained generation of free radicals and non radical species derived from free radicals can lead to an imbalance in the intracellular redox homeostasis. Normally, any excess levels of reactive radical and nonradical species generated are intercepted by antioxid...
متن کاملO-21: Differential Expression and Epigenetic Pattern of HOX Family Genes in Cumulus Cells of Mature MII Oocytes from Patients with Polycystic Ovary Syndrome
Background Ovarian tissue cryopreservation represents a promising strategy to preserve the ovarian function in cancer patients. It is usually performed by slow freezing/rapid thawing (SF/RT). Recent studies emphasize an ultrarapid cryopreservation procedure, vitrification/warming (V/W), since it might prevent damages due to ice crystal formation. Comparative studies between the cryopreservation...
